☞ Plasmid DNA를 restriction enzyme으로 자른 후 DNA size marker와 비교하여 본다. Cleavage는 enzyme이 인식하는 특정한 recognition sequence로부터 >1,000 b.p 떨어진 non-specific site에서 일어난다.

☞ Restriction enzyme digestion
